Amyotrophic Lateral Sclerosis

The Problem

ALS, also known as Lou Gehrig's disease, is a fatal neurodegenerative disorder characterized by the progressive loss of motor function. Nerve cells that control muscle function are lost in ALS patients.

Asha's Solution

SARM1 is a key regulator of nerve cell degeneration in ALS. Asha has developed a novel intramolecular glue inhibitor of SARM1 that is restoring motor function in preclinical models of ALS.

Alzheimer's Disease

The Problem

AD is characterized by the loss of cognitive function and progressive dementia. Deposition of protein aggregates in the brain of AD patients leads to mitochondrial dysfunction and cell death pathway activation, resulting in neurodegeneration.

Asha's Solution

Mitochondrial fragmentation in the AD brain drives neurodegeneration. Activation of a key protein that controls mitochondrial homeostasis, DRP1 is altered in patients. Asha has developed a novel and selective DRP1 inhibitor that restores normal mitochondrial homeostasis.

Parkinson's Disease

The Problem

PD is a chronic and progressive neurodegenerative disroder that results from the loss of dopaminergic neurons leading to both motor and non-motor symptoms.

Asha's Solution

PD pathology is highly linked to mitochondrial dysfunction. A variety of pathways lead to mitochondrial dysfunction in PD, including DRP1-mediated mitochondrial fragmentation. Asha's DRP1 inhibitor shows significant promise as a disease-modifying therapeutic for PD.

Chemo-induced Peripheral Neuropathy

The Problem

CIPN affects over 80% of cancer patients taking platinum-based and taxane chemotherapeutics, often resulting in dose-limiting neuropathy. Overcoming this neuropathy provides a path to increased chemo dosing & better patient response.

Asha's Solution

Like in ALS, SARM1 is activated in CIPN, resulting in nerve degeneration and neuropathic pain. Preclinical studies using Asha's SARM1 inhibitor show a near complete amelioration in pain symptoms during chemo dosing.
